Combining insect genitalia preparation with DNAextraction

Dear colleagues,

We (Knölke et al. ) developed the method of combined DNA extraction and
gentitalia preparation in the course of the DNA-TAX - project of BIOLOG,
a framework project on biodiversity informatics of the Geman Federal
Ministry of Science (BMBF).

The exact protocoll will be published in "Insect Systematics and
Evolution" Vol. 35(4) early next year, and we will announce the out of
press status via TAXACOM.

In addition to Lepidoptera, we applied this method also for Odonata and
Blattoptera with convincing results. Especially in new species
descriptions and larvae assignments it will be the outstanding technique
to combine genetic and morphological investigations. Our long term
experience shows that there are no restrictions of the method, but
certain adaptations are required in some cases.

Yes, I have done 10 th's this way, mostly of very small moths (Nepticulidae).
The protocol was developed by Sonja Knölke, Andreas Segerer and others in Munich. A paper (Knölke et al) is in press in Insect Systematics and Evolution, as far as I know not yet published. They suggest that this should be standard procedure!

The results are good: enough DNA extracted and perfectly clean genitalia!

Hello All,

Has anyone experimented with using a proteinase-K/buffer solution to clear insect abdomens for genitalia preparation, rather than using 10% KOH?  Seems to me this would allow you to extract DNA from the gunk that one normally throws out...  If you have tried this yourself or know of references please e-mail me.
