If by "American" you include North and South America, then yes,
collectors do have an impact on Theraphosidae.  Witness EURYPELMA SMITHI,
the Mexican Red-Legged (or Red-Kneed) tarantual, the only spider listed
in CITES.

In the U.S. of A., in Texas in particular, I believe that more members of
this family are lost by being squished on the highways than collectors
ever thought of collecting.
